In 2019, a plague has transformed 90% of the world's human population into vampires. As the vampires are incapable of aging or dying, but they are unprotected against the UV lights or the sunlight, which is why the whole vampire world is active at night. Human numbers dwindle and the need for blood becomes desperate while an all-out war rages around the world between the surviving humans and vampires despite their efforts. When deprived of blood for extended periods, vampires degenerate into subsiders: aggressive bat-like creatures. Humans are captured and harvested in laboratory farms while scientists research a synthetic blood substitute to satisfy vampires' blood hunger. The main supplier of blood in the United States is the pharmaceutical company Bromley Marks, run by Charles Bromley (Sam Neill). In 1961, the Ark, a Cybertronian spacecraft carrying an invention capable of ending the war between the philanthropic Autobots and the malevolent Decepticons, crash lands on the dark side of Earth's Moon. The crash is detected on Earth by NASA, and President John F. Kennedy authorizes a mission to put a man on the Moon as a cover for investigating the craft. In 1969, the crew of Apollo 11 lands on the Moon.
In the present, the Autobots assist the United States military in preventing conflicts around the globe. During a mission to Chernobyl, to investigate suspected alien technology, Optimus Prime finds a fuel cell from the Ark, discovering that it had survived its journey from Cybertron. The Autobots are attacked by Shockwave, who manages to escape. After learning of the top-secret mission to the Moon, the Autobots travel there to explore the Ark. They discover a comatose Sentinel Prime – Optimus' predecessor as leader of the Autobots – and the Pillars he created as a means of establishing a Space Bridge between two points to teleport matter. After returning to Earth, Optimus uses the energy of his Matrix of Leadership to revive Sentinel Prime.

In the late Third Age, Sauron's power is increasing, and he has sent his Orcs to seek out the remnants of the bloodline of Elendil, kept alive in the Dúnedain. Dirhael, his wife Ivorwen and their daughter Gilraen are fleeing from an attack on their village when they are ambushed by Orcs on a forest road, and saved by a group of rangers led by Arathorn. Not having any place safer to go, the refugees go with Arathorn to Taurdal, the village led by his father and Chieftain of the Dúnedain, Arador. While there, Arathorn and Arador ponder the Orcs' motives after finding various pieces of jewelry on their bodies. During her stay in Taurdal, Gilraen falls in love with Arathorn.In light of the attacks on surrounding settlements, Arador leads his forces on a campaign against the Orcs in the area in an attempt to restore peace to the region. Meanwhile, he sends Arathorn separately in an attempt to determine the meaning behind the attacks. Both are successful, and Arathorn discovers the orcs are serving Sauron, who seeks the Ring of Barahir. Arathorn and Gilraen receive Arador's blessing to be wed, but Arathorn cannot summon the courage to ask Dirhael for his daughter's hand. Arador is summoned to Rivendell to seek Elrond's council, and the wedding is postponed until his return. Arathorn eventually confronts Dirhael, and receives permission to marry his daughter. Arathorn and Gilraen are married.

The movie opens with Elise (Angelina Jolie) being followed by French Police, working with Scotland Yard under the direction of Inspector John Acheson (Paul Bettany). Acheson has spent years hunting Elise's old lover, Alexander Pearce, who owes £744 million in back taxes and is believed to have received 20 million dollars worth of plastic surgery to alter his appearance completely. At a cafe, Elise receives written instructions from Pearce: board a train to Venice, pick out a man who resembles Pearce, and make the police believe that this decoy is Pearce himself. Elise burns the note, then manages to evade the police and board the train. On the train, Elise picks Frank (Johnny Depp), an American high school teacher. She spends much time with him, seeming to start a romance. In the meanwhile, the police have managed to salvage the ashes of her burned note and assembled them to extract information regarding her rendezvous as well as her ruse. But an informer from the police station mistakenly communicates to Reginald Shaw (Steven Berkoff), a gangster from whom Pearce as accountant stole $2.3 billion, that Pearce is travelling with Elise on the train to Venice. Shaw immediately proceeds to Venice.Elise invites Frank to stay with her at the hotel room that has been arranged for her in Venice. Pearce leaves further instructions for Elise to attend a ball. Elise abandons Frank, who is then chased by Shaw's men. While trying to escape from them, Frank is detained by the Italian police ostensibly for his own safety, only to have a corrupt inspector turn him over to Shaw's men in exchange for the bounty that has been placed on Pearce's head. Elise rescues Frank just before he is handed over, leading Shaw's men on an extended boat chase and escaping. She leaves Frank at the airport with his passport and money, urging him to go home for his own safety.
